Human milk oligosaccharides (HMOS) are the third most abundant solid component in human milk. Their levels vary with maternal diet and ethnicity. In a sample of breast milk, they reach 5-20 g/L, depending on lactation stage and parity. HMOS are a complex group of glycolipids that are produced by a variety of Le-gene products. However, the relative amounts of HMOS in samples are not well known. Therefore, this study aimed to quantify HMOS and describe their relative levels.
HMOS were determined by highly sensitive multiplexed capillary-gel electrophoresis with laser-induced fluorescence detection. The presence of HMOS in samples was categorized into a few groups. One of the groups was characterized by a high concentration of b3′-GL.
Another group was defined by a high concentration of a’-GL. The remaining groups were characterized by low levels of GLs. These GLs occur in a series of structurally distinct isomers. For example, b3′-GL has been shown to inhibit the toll-like receptor signaling in immature human intestinal cells.
